Analysis of the Experimental Lesion of Connective Tissue Produced by a Complex of C Polysaccharide from Group a Streptococci

چکیده
Rabbits presumably sensitized to components of the streptococcal cell by natural contact with cross-reactive microbial antigens in their environment, display distinctive phases of evolving allergic response to an extract of streptococcal cells. Animals under 4 weeks of age show no gross allergic skin reaction; this is followed successively by age periods in which allergic reactions become most prominent at 4 hours, 23 hours, or 2 to 4 days after injection. A relapsing nodular lesion, produced by cell wall components in the cell extract, occurs in neonatal rabbits which display no hypersensitivity, reaches a peak response in young animals during the period of most active growth, and decreases in severity in older rabbits while the degree of hypersensitivity is increasing. Relapses occur with greater frequency in neonatal than in older rabbits. Passive transfer of spleen cells or serum from sensitized adult rabbits has no influence on the nodular reaction in neonatal animals.

Immunological Studies on a C Polysaccharide Complex of Group a Streptococci Having a Direct Toxic Effect on Connective Tissue
A macromolecular complex of C polysaccharide and peptide derived from Group A streptococcal cells produces a multinodular, remittent, and intermittent lesion of dermal connective tissue following a single intradermal injection. This lesion is greatly modified, both in the gross and microscopically, in rabbits hyperimmunized with a Group A streptococcal vaccine. The immune serum, absorbed to yie...
متن کاملRelation of Particle Size of C Polysaccharide Complexes of Group a Streptococci to Toxic Effects on Connective Tissue
The component of Group A streptococci which is responsible for the chronic, remittent, multinodular lesion of connective tissue is derived from the cell wall. Further evidence is given to support the essential role of the group-specific C polysaccharide in the production of this lesion. A series of particles containing the group-specific C polysaccharide was prepared, ranging in size from large...
متن کاملIdentification of a Toxic Cellular Component of Group a Streptococci as a Complex of Group-specific C Polysaccharide and a Protein
The toxic cellular component of Group A streptococci, which produces a chronic multinodular lesion of the dermal connective tissue of rabbits, has been isolated by means of differential centrifugation and electrophoresis. The substance appeared to be a macromolecular complex of polysaccharide and protein, the polysaccharide having the serological and chemical characteristics of the group-specif...
متن کاملToxic Cellular Component of Group a Streptococci as a Complex of Group- Specific C Polysaccharide and a Protein* by John H. Schwab, Ph.d.,
Chronic multinodular skin lesions were produced in rabbits injected intradermally at a single site with a sterile extract of sonically disrupted Group A streptococcal cells (1). These lesions, which consisted of separate nodules scattered over areas as large as 12 cm. in diameter, presented several remarkable features. The injury to dermal connective tissue was evident within 48 hours after inj...
متن کاملPersistence of Group a Streptococcal Cell Walls Related to Chronic Inflammation of Rabbit Dermal Connective Tissue
Antibodies specific for the mucopeptide and group-specific C polysaccharide antigens of Group A streptococcal cell walls were prepared by acid dissociation of immune precipitates, and labeled with either fluorescein or (125)I. Employing both fluorescent and radioautographic procedures the persistence of the antigens was followed in skin sites injected with cell wall fragments. Both antigens per...
